اثر روش خاکورزی و مدیریت بقایای گندم (Triticum aestivum L.) بر عملکرد ذرت (Zea mays L.)، خصوصیات خاک و فراوانی کرمهای خاکی
روش خاکورزی و مدیریت بقایا دو عامل مهم مدیریتی هستند که میتوانند ویژگیهای محیط و موجودات خاک و تولید زراعی را تحت تأثیر قرار دهند. آزمایشی سه ساله به منظور بررسی اثر خاکورزی (خاکورزی رایج، کم خاکورزی و بی خاکورزی) و مدیریت بقایای گیاهی گندم (حفظ بقایا و حذف تمام بقایا از سطح خاک) بر فراوانی کرمهای خاکی، ماده آلی خاک، نفوذ تجمعی و عملکرد ذرت، به صورت کرتهای یک بار خُرد شده با سه تکرار در زرقان فارس اجرا شد. بیشترین عملکرد دانه (14447 کیلوگرم در هکتار) و شاخص برداشت ذرت (82/36 درصد) به ترتیب در کم خاکورزی و خاکورزی رایج با حفظ بقایای گیاهی حاصل شد. عملیات کم خاکورزی و حفظ بقایا میزان عملکرد دانه ذرت را نسبت به بی خاکورزی بیش از 30 درصد افزایش داد. خاکورزی رایج و حذف بقایا نسبت به خاکورزی حفاظتی منجر به کاهش 15 و 12 درصدی ماده آلی خاک در دو عمق صفر-10 و 10-20 سانتیمتری شد. با حفظ بقایای گیاهی بیشترین میزان نفوذ تجمعی در کم خاکورزی و خاکورزی رایج حاصل شد. بیشترین فراوانی کرمهای خاکی در بی خاکورزی و حفظ بقایا حاصل شد در حالی که خاکورزی رایج و حذف بقایا نسبت به کم خاکورزی و بی خاکورزی فراوانی کرمهای خاکی را به ترتیب 5/38 و 50 درصد کاهش داد. بهطور کلی در تناوب پیوسته گندم-ذرت جهت حفظ پتانسیل عملکرد ذرت، بهبود حاصلخیزی خاک و افزایش فعالیت کرمهای خاکی انجام عملیات کم خاکورزی و حفظ بقایای ایستاده گیاهی گندم با ارتفاع 30 سانتیمتر در سطح خاک قابل توصیه است.

جداسازی، شناسایی و بررسی ویژگیهای محرک رشدی سودوموناسهای فلورسنت از ریزوسفر درختان زیتون در خاکهای شور
سودوموناسها گروه مهمی از باکتریهای ریزوسفری محرک رشد هستند که به دلیل توزیع گسترده در خاک، توانایی کلونیزاسیون ریزوسفر بسیاری از گیاهان و دارا بودن ویژگیهای محرک رشدی متعدد از اهمیت ویژهای برخوردار هستند. پژوهش حاضر به منظور شناسایی جدایههای سودوموناسهای فلورسنت از ریزوسفر درختان زیتون در باغهای شور استان قزوین و ارزیابی خصوصیات محرک رشدی این جدایهها از قبیل توانایی تولید سیدروفور، اکسین، سیانید هیدروژن، آنزیم ACC deaminase، قابلیت انحلال فسفر در محیط مایع، تغییرات pH و میزان تحمل غلظتهای مختلف نمک (100، 200 و 400 میلیمولار) انجام شد.به این منظور 15 جدایه سودوموناس از42 نمونه خاک ریزوسفری از باغات زیتون استان قزوین جمعآوری گردید. بر اساس نتایج بالاترین مقدار تولید آنزیم ACC deaminase در جدایههای 7Q و 9Q مشاهده شد. بیشترین مقدار تولید اکسین در حضور ppm100 از ال-تریپتوفان مربوط به جدایه 2Q (96/11میکرو گرم بر میلیلیتر بود. انحلالپذیری تریکلسیم فسفات در محیط مایع در ارتباط با pH همبستگی منفی نشان داد و بالاترین مقدار فسفر حل شده برابر با 35/251 میکروگرم بر میلیلیتر متعلق به جدایه 13Q بود. همچنین 40% جدایهها قادر به تولید HCN نبودند. از نظر تولید سیدروفور در دورههای زمانی دو، چهار و شش روزه تمامی جدایهها قادر به رشد و تولید سیدروفور در محیط CAS-Agar بودند. همچنین تمامی جدایهها به استثنای جدایههای 1Q و 8Q در غلظتهای مختلف نمک به خوبی رشد کردند. باتوجهبهنتایجحاصل از این پژوهش و پتانسیل شناسایی شده در این جدایهها، برخی از این جدایهها میتوانند به عنوان کاندیدا جهت انجام مطالعات ژنومیکس و همچنین افزایش تحمل به شوری گیاهان در باغات شور مورد استفاده قرار گیرند.

اثر برخی ریز جانداران در کاهش آلودگی یک خاک آهکی آلوده به نفت خام
آزمایشی گلخانهای به صورت فاکتوریل در قالب طرح کاملاً تصادفی به منظور بررسی اثر تیمارهای زیستی (باکتری سودوموناس فلورسنس، باکتری میکروکوکوس یوننانسیس، قارچ کلارودیوگلوموس اتونیکاتوم و قارچ فونلیفورمیس موسه) بر پاسخهای گیاه وتیور در آلودگی نفت خام (0، 2 و 4 درصد وزنی) انجام شد. نتایج نشان داد افزایش سطح آلودگی نفتی وزن خشک اندام هوایی و ریشه و همچنین درصد کلنیزاسیون ریشه را کاهش داد اما تنفس میکروبی و غلظت کل هیدروکربنها در خاک پس از برداشت را افزایش داد. تلقیح باکتری میکروکوکوس یوننانسیس و قارچ فونلیفورمیس موسه وزن خشک اندام هوایی را افزایش داد، همه تیمارهای بیولوژیکی بجز باکتری سودوموناس فلورسنس افزایش وزن خشک ریشه را موجب گردیدند. مایه زنی قارچ کلارودیوگلوموس اتونیکاتوم باعث افزایش معنیدار غلظت و جذب آهن، و قارچ فونلیفورمیس موسه باعث افزایش معنیدار میانگین غلظت روی و جذب روی و آهن شد. همچنین مایه زنی قارچ باعث افزایش درصد کلنیزاسیون ریشه شدند. از بین ریزجانداران مطالعه شده باکتری میکروکوکوس یوننانسیس و قارچ فونلیفورمیس موسه تنفس میکروبی را افزایش دادند که به نوبه خود مقدارکل هیدروکربنهای باقی مانده در خاک پس از برداشت را کاهش داد. نتایج ما نشان داد که از بین ریزجانداران باکتری میکروکوکوس یوننانسیس و قارچ فونلیفورمیس موسه اثرات مطلوبتری در زیست پالایی و مقاومت گیاه به آلودگی نفتی دارند.

بررسی نوسان جمعیت و مدل مطلوبیت زیستگاه دمفنریان راسته Entomobryomorpha (Collembola, Hexapoda) در زیستگاههای مختلف استان کرمانشاه
در این تحقیق جمعیت دمفنریان راسته Entomobryomorpha در سه اکوسیستم مختلف: زمین زراعی، جنگل بلوط و مراتع سه شهرستان اسلامآباد غرب، پاوه و سرپلذهاب مورد ارزیابی قرار گرفت. نمونهبرداری ماهیانه، از خرداد 1395 تا اردیبهشت 1397 صورت پذیرفت. از هر ایستگاه 10 نمونه خاک به سطح 100 سانتیمتر مربع و عمق 13 سانتیمتر تهیه و جمعیت دمفنریان شمارش شد. همزمان، دما و رطوبت خاک نیز ثبت گردید. جهت محاسبه مطلوبیت زیستگاه، با استفاده از روش رگرسیون لوجستیک ارتباط بین حضور و عدم حضور گونه و شرایط زیستگاه از نظر رطوبت و دما مقایسه شد. بطور کلی از مجموع 16 گونه شناخته شده، زیستگاه جنگل نسبت به دو زیستگاه دیگر دارای تعداد گونه بیشتری بود (به طور میانگین %2/79 از کل گونهها). از سوی دیگر در شهرستان اسلامآباد غرب و سرپلذهاب، بیشترین درصد فراوانی دمفنریان، در جنگل (به ترتیب با 77/60 و 22/69 درصد) بوده و در شهرستان پاوه مرتع (% 19/49) از دو زیستگاه دیگر درصد فراوانی بیشتر داشت. نتایج شمارش ماهیانه و بررسی ارتباط بین حضور و عدم حضور گونهها بر اساس دما و رطوبت نشان داد فعالیت اکثر گونهها بین ماههای آبان تا اردیبهشت است. حضور اکثر گونهها ارتباط مستقیمی با افزایش رطوبت و رابطه عکسی با دمای خاک داشت اما، برخی گونهها مانندSeira sp. در رطوبتهای زیر ده درصد (رطوبت شش درصد مزرعه و هشت درصد مرتع) و دمای بالای خاک (35 تا 40 درجه سلیسیوس) فعالیت داشتند. این امر نشان میدهد علاوه بر دو فاکتور دما و خاک، فاکتورهای دیگری نیز در این امر دخالت دارند.

شناسایی و بررسی خصوصیات محرک رشدی و بیوکنترلی قارچهای اندوفیت جدا شده از برگ و میوه پسته
قارچهای اندوفیت با سازوکارهای مختلف باعث افزایش رشد گیاه و همچنین مقاومت آن به تنشهای زنده و غیرزنده میشوند. به این منظور جهت شناسایی قارچهای اندوفیت درخت پسته و ارزیابی ویژگیهای محرک رشدی و بیوکنترلی آنها، از برگ و میوه درختان سالم پسته شهرستان رفسنجان نمونهبرداری انجام گرفت. پس از شناسایی ریختشناسی و مولکولی، فاکتورهای محرک رشدی از قبیل تولید اکسین، سیدروفور، حل کنندگی فسفات معدنی و فعالیت بیوکنترلی این قارچها از طریق تولید کیتیناز و سیانید هیدروژن ارزیابی گردید. مقایسه میانگین دادهها نشان داد که بیشترین میزان تولید اکسین در سطح صفر و 50 میلیگرم در لیتر تریپتوفان به ترتیب به میزان 34/19 و 88/32 میلیگرم در لیتر توسطTrichoderma atroviride TA2-2-1 تولید گردید. بیشترین میزان سیدروفور توسط Quambalaria cyanescens QC11-3-2 تولید شد که نسبت قطر هاله به پرگنه 96/2 بود. تنهاByssochlamys nivea BN1-1-1 قابلیت تولید حل کنندگی فسفات معدنی را داشت که نسبت قطر هاله به پرگنه 1/1 بود. بیشترین فعالیت کیتینازی به ترتیب در جدایه TH5-1-2 Trichoderma harzianum (U/mL 92/2) وT. atroviride TA2-2-1 (U/mL 34/2) مشاهده شد. گونههایT. atroviride TA2-2-1 و Clonostachys rosea CR2-3-1 قادر به تولید به مقدار زیاد سیانید هیدروژن (درجه 5) بودند.

اثر مایهزنی باکتری محرک رشد گیاه و سطوح رطوبتی مختلف بر شکلهای شیمیایی کادمیم در خاک پس از برداشت ذرت
کادمیم به عنوان یکی از خطرناکترین عناصر برای جانداران شناخته میشود و به دلیل قابلیت دسترسی آسان برای گیاه، یک تهدید بزرگ برای چرخه غذایی محسوب میشود. بمنظور بررسی اثر افزودن کودهای بیولوژیک همچون باکتری محرک رشد گیاه میکروکوکوس یونانیسیس بر شکلهای شیمیایی این فلز سنگین در خاک، آزمایشی گلخانهای بصورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار انجام شد. تیمارها شامل دو سطح باکتری (با و بدون مایهزنی باکتری)، چهار سطح کادمیم (شاهد، ١٠، ٢٠، و٤٠ میلیگرم در کیلوگرم خاک)، و سه سطح رطوبتی مختلف (100، 80، و 65 درصد رطوبت ظرفیت مزرعه) بود.پس از برداشت ذرت، شکلهای شیمیایی کادمیم در خاک بررسی گردید. نتایج نشان داد که با افزایش سطوح کادمیم کاربردی، مقادیر شکلهای شیمیایی مختلف کادمیم نیز افزایش یافت. مایهزنی باکتری سبب کاهش شکلهای تبادلی، کربناتی، آلی، متصل به اکسیدهای آهن و منگنز شد اما شکل باقیمانده با مایهزنی باکتری افزایش یافت و کادمیم به شکل کم محلولتر تبدیل شد. با کاهش رطوبت خاک، در شرایط بدون مایهزنی شکلهای آلی و متصل به اکسیدهای آهن بیشکل کاهش و در شرایط مایهزنی شده شکلهای متصل به اکسیدهای آهن بیشکل و منگنز افزایش یافت. بیشترین مقدار نسبی کادمیم مربوط به شکل کربناتی و سپس به ترتیب شکلهای باقیمانده > متصل به اکسیدهای منگنز > متصل به اکسیدهای آهن بیشکل > آلی > تبادلی بود.

بهرهگیری از ریزسازوارههای خاکزی میتواند بخشی از اثرات منفی تنش شوری را کاهش دهد. بهمنظور بررسی اثر شوری، قارچ میکوریزا آربوسکولار و باکتری حل کننده ترکیبات فسفری بر خصوصیات رشدی گیاه دارویی اسفرزه (Plantagoovata Forsk.) دو آزمایش طراحی و اجرا شد. در آزمایش اول بهمنظور غربالگری گونه مقاوم باکتری حلکننده فسفات در شرایط تنش شوری، 40 باکتری تحت آزمون نیمه کمی توان انحلال فسفات در شرایط شوری محیط کشت قرار گرفتند. برای این ارزیابی میزان نسبی انحلال فسفات، نسبت قطر هاله بر قطر کلنی تعیین گردید. سپس گونه برتر که بالاترین توان حل فسفات معدنی در شوری 60 دسی زیمنس بر متر داشت، انتخاب شد. مقایسه توالی 16S rRNA و آنالیز فیلوژنتیکی نشان داد که سوش جدا شده Pseudomonas fluorescens بود. آزمایش دوم بهصورت فاکتوریل سه عاملی در قالب طرح بلوکهای کامل تصادفی با سه تکرار بود. عامل اول سه سطح شوری شامل 5/2 ، 5 و 10 دسیزیمنس بر متر، عامل دوم قارچ میکوریزا شامل عدم کاربرد قارچ و گونههای Funneliformis mosseae، Rhizophagus intraradices و Glomus fasciculatum و عامل سوم شامل دو سطح عدم کاربرد باکتری و کاربرد باکتری برتر حاصل از آزمایش شناسایی باکتری بود. صفات مورد اندازهگیری شامل وزن خشک ساقه و ریشه، نسبت وزن خشک ریشه به ساقه، پاسخ رشد میکوریزایی و درصد کلونیزاسیون ریشه بود. نتایج تجزیه واریانس نشان داد اثر متقابل تنش شوری و قارچ میکوریزا بر صفات وزن خشک ساقه و ریشه، نسبت وزن خشک ریشه به ساقه، پاسخ رشد میکوریزایی و درصد کلونیزاسیون ریشه و اثر متقابل تنش شوری، قارچ میکوریزا و باکتری بر وزن خشک ساقه در سطح احتمال ۱ درصد معنیدار بود. همچنین اثر متقابل تنش شوری و باکتری بر نسبت وزن خشک ریشه به ساقه در سطح احتمال ۵ درصد معنیدار بود. بیشترین وزن خشک ریشه و نسبت وزن خشک ریشه به ساقه در ترکیب تیماری 5/2 دسی زیمنس بر متر و کاربرد قارچ Glomus fasciculatum بهمیزان 07/1 گرم در گیاه و 29/0 حاصل شد. بیشترین پاسخ رشد میکوریزایی در ترکیب تیماری شوری 10 دسی زیمنس بر متر + قارچ Rhizophagus intraradices بهمیزان 7/76 درصد بود. مقایسه میانگین اثر متقابل تنش شوری و باکتری نشان داد بیشترین پاسخ رشد میکوریزایی در ترکیب تیماری شوری 10 دسی زیمنس بر متر + کاربرد باکتری Pseudomonas fluorescens بهمیزان 6/45 درصد بود و بیشترین کلونیزاسیون ریشه در شوری 10 دسی زیمنس بر متر بهوسیله قارچ میکوریزا Rhizophagus intraradices بهمیزان 1/65 درصد حاصل شد. نتایج نشان داد هر چند افزایش شوری توانست مولفههای رشدی گیاه را کاهش دهد، لکن کاربرد همزمان باکتری حل کننده فسفات معدنی و قارچ میکوریزا توانست اثرات منفی تنش شوری را تا حدی جبران نماید. با توجه به نتایج حاصل از این پژوهش میتوان کاربرد همزمان باکتری Pseudomonas fluorescens و قارچ Rhizophagus intraradices را برای دستیابی به حداکثر تولید گیاه دارویی اسفرزه توصیه نمود.

اثر تنظیم pH کمپوست شهری بر غنیسازی آن با باکتری محرک رشد گیاه Enterobacter cloacae
برای افزایش کارایی و کیفیت کمپوست، غنیسازی عنصری و میکروبی از راهکارهای اساسی است. افزودن باکتریهای مفید و محرک رشد گیاهان از جنبههای مختلف زیستمحیطی مورد تأیید میباشد. از عوامل مؤثر بر مدت زندمانی باکتری در کمپوست، pH آن میباشد. در این تحقیق کمپوست شهری را با اتوکلاو استریل نموده و به دو بخش شامل الف- کمپوست با pH اولیه (6/5) و ب- کمپوست با pH تنظیم شده در هفت توسط کربنات کلسیم) تقسیم شد. مایهزنی با باکتری Enterobacter cloacae با جمعیت CFU g-1 109 انجام گرفت. رطوبت کمپوست در حدود 40 درصد ظرفیت نگهداری آب تنظیم و نمونهها در انکوباتور با دمای 26 درجه سلسیوس نگهداری شدند. شمارش جمعیت میکروبی و اندازهگیری pH کمپوست در زمانهای 3، 15، 45، 75، 105، 135 ، 165 و 195روز پس از تلقیح انجام گردید. نتایج شمارش جمعیت میکروبی نشان داد که اثر pH در زندهمانی باکتری در کمپوست معنیدار بود (p<0.01). 45 روز پس از تلقیح، جمعیت میکروبی در کمپوست باpH تنظیم نشده به حدود CFU g-1 104 تقلیل یافت در حالی که در کمپوست تنظیم شده از نظر pH، این تعداد برابر CFU g-1 109 بود. علیرغم نوسانات جمعیت میکروبی در کمپوست تنظیم نشده، تا 135 روز بعد از تلقیح در هر دو کمپوست جمعیت CFU g-1 107 حاکم بود. تعداد جمعیت باکتری در کمپوست تنظیم شده بعد از 165 روز و کمپوست بدون تنظیم بعد از 140 روز به جمعیت زیر حد استاندارد (CFU g-1 106) رسیدند.
