بررسی فعالیت و پایداری آنزیم لاکاز آزاد تثبیت شده در حضور کانیهای مونتموریلونیت و زئولیت
آنزیم لاکاز از گروه مهمترین آنزیمهایی است که در سالهای گذشته به سبب توانایی آن در اکسید کردن ترکیبهای گوناگون و آلایندههای محیطی پایدار، نگاه بسیاری را در زمینه زیست بهسازی به سوی خود جلب کرده است. با نگاه به پایداری کم و هزینههای ساخت و فراوری بالای آنزیمها، بیجنبسازی آنها بر رویه نگهدارندهها میتواند راهی شایسته در راستای افزایش پایداری کارکرد آنزیم باشد. برای بررسی برهمکنش لاکاز گرفته شده از قارچ ترامتس ورسیکالر (Trametes versicolor) با کانیهای مونتموریلونیت و زئولیت سه آزمایش جداگانه در قالب طرح کاملاً تصادفی با دو تکرار در شرایط آزمایشگاهی انجام شد. آزمایشها به ترتیب شامل چهار سطح pH (5، 6، 7 و 8)، نه سطح دمایی (4 و80-10 درجه سانتیگراد) و هفت سطح زمان انکوباسیون (صفر، 1، 2، 5، 10، 20 و 30 روز) بودند. بر پایه آزمایش نخست، بالاترین اندازه جذب آنزیم بر رویههای کانیهای بررسی شده در 5=pH دیده شد و با افزایش pH، جذب آنزیمی کاهش یافت. در آزمایش دوم، بالاترین اندازه فعالیت نسبی برای لاکاز آزاد در دمای 20 درجه سانتیگراد، برای لاکاز بیجنبشده بر مونتموریلونیت در دماهای 80 و 4 درجهسانتیگراد و برای لاکاز بیجنبششده بر زئولیت در دماهای 4 و 70 درجهسانتیگراد بود. لاکاز بی جنبششده پایداری خوبی را در برابر دماهای پایین و بالا نشان داد. در آزمایش سوم، بالاترین اندازه فعالیت نسبی (100 درصد) برای لاکاز بیجنبششده بر مونتموریلونیت و زئولیت در زمان انکوباسیون 20 روز و برای لاکاز آزاد در زمان 5 روز دیده شد. برپایه این پژوهش، بیجنبشسازی پیامد شایستهای را بر پایداری فعالیت آنزیم لاکاز نشان داد.

تعیین مشخصات باکتریهای ریزوبیا جدا شده از خاکهای آلوده به آرسنیک در مناطق جنوب شرقی استان کردستان و تأثیر آنها بر روی رشد گیاه
باکتریهای همزیست با ریشه بقولات از ریشه گیاهان یونجه (Medicago sativa L.)، نخود (Cicer arietinum L.) و یونجه زرد (Melilotus officinalis L.) رشد کرده در خاکهای آلوده به آرسنیک در جنوب شرقی استان کردستان جداسازی گردید. جدایهها براساس خصوصیات فیزیولوژیکی و بیوشیمیایی متعلق به جنس Rhizobium و Ensifer تشخیص داده شد. وجود سیستم مقاومتی به آرسنیک، سیستم ars، با استفاده از آغازگرهای اختصاصی ژن arsC بررسی و به اثبات رسید. نرخ رشد جدایهها در محیط کشت YEMB به اضافه غلظتهای 400 – 100 میلی مولار آرسنات بررسی گردید. سویههای جدا شده از یونجه (AB1، AB3)، نخود (PA2، PC2) و یونجه زرد (YA1) توانائی تحمل 350 میلی مولار آرسنات را داشتند. همچنین سویههای AB1، PA2 و YA1 توانائی تحمل غلظت 400 میلی مولار آرسنات را داشتند. به منظور بررسی تأثیر آلودگی به آرسنیک بر روی رابطه همزیستی یونجه و نخود با باکتریهای ریزوبیا و میزان رشد گیاهان، آزمونی در قالب بلوکهای کاملاً تصادفی در سه تکرار و در پنج غلظت آرسنیک (100، 75، 50، 10 و صفر میلیگرم بر کیلوگرم خاک) و با تلقیح یا بدون تلقیح دو سویه باکتری، AB1 و PA2 انجام گردید. وزن خشک و وزن تر ریشه و اندامهای هوائی یونجه و نخود با افزایش غلظت آرسنیک در خاک کاهش یافت. نتایج نشان داد که وزن خشک و وزن تر ریشه و اندامهای هوائی هر دو گیاه در بوتههای تیمار شده توسط سویههای باکتری بصورت معناداری بالاتر از فاکتورهای مشابه در گیاهان تلقیح نشده با باکتری بود. براساس نتایج بدست آمده سویههای AB1 و PA2 با توانائی مقاومت به غلظتهای بالای آرسنیک به همراه گیاهان همزیست میتوانند جهت پالایش خاکهای آلوده به آرسنیک مورد استفاده قرار گیرند.

بررسی افزایش وزن و برخی شاخصهای رشد کرم خاکی در ورمی کمپوست حاصل از کودهای آلی مختلف
تهیه ورمی کمپوست یکی از مهمترین ابزارها برای بازیافت کودهای آلی و تبدیل آنها به کود بیولوژیک است. بررسی مورد نظر در جهت تعیین بهترین بستر جهت پرورش کرم خاکی گونه آیزینا فتیدا (Eisenia .foetida) در یک طرح کاملاً تصادفی در10 تیمار و چهار تکرار در یک دوره سه ماهه در بسترهای مختلف کود دامی اسبی و گوسفندی به همراه بقایای کاه گندم و یونجه در نسبتهای حجمی مختلف (25، 50، 75 و 100) در سال 1392 در دانشگاه گنبد کاووس انجام شد (تیمارهای کودی گنگ است نسبتها ذکر نشده است). در این تحقیق وزن کرم بالغ، جوان، نوزاد، وزن کوکون و وزن نهایی کرم خاکی آیزنیا فتیدا و نرخ رشد وزنی، نرخ رشد ویژه تعداد نوزاد به ازای مولد و نرخ بازماندگی مورد بررسی قرار گرفت. نتایج حاصل از بررسی تجزیه واریانس نشان داد که بین تیمارها از نظر افزایش وزن کرم بالغ، جوان، نوزاد و وزن کرم نهایی و شاخصهای رشد غیر از نرخ بازماندگی اختلاف معنیدار وجود داشت. بیشترین وزن کرم بالغ در تیمار کود گوسفندی و بقایای یونجه به نسبت 50 به 50 (نسبت حجمی) مشاهده گردید و بیشترین وزن کرم جوان در تیمار کاه به همراه کود گوسفندی به نسبت 25 به 75 حاصل شد. همچنین بیشترین وزن نوزاد کرم و وزن نهایی کرمها در تیمار کاه به همراه کود گوسفندی به نسبت یک به یک به دست آمد. در مجموع نتایج حاصل نشان داد که تیمار کود گوسفندی به همراه بقایای کاه و یونجه بیشترین افزایش وزن کرم خاکی را به دنبال داشت.

جداسازی و شناسایی باکتریهای مولد اگزو پلی ساکارید بومی خاکهای شور
اگزو پلی ساکاریدهای مترشحه از باکتریها دارای نقش مهمی در مقاومت باکتریها در مقابل تنشها از جمله شوری میباشند. این مطالعه با هدف جداسازی و شناسایی باکتریهای شورزی با بیشترین توان تولید اگزو پلی ساکارید از خاک و بررسی میزان تولید اگزو پلی ساکارید در غلظت های مختلف نمک انجام گردید. نمونههای خاک مورد نظر بر محیط نوترینت آگار دارای 5 % نمک کشت شد. از میان باکتریهای شورزی، کلونیهای موکوئیدی با قدرت رشد در حضور بیشترین غلظت نمک به عنوان کلونی مولد اگزو پلی ساکارید انتخاب و مقدار اگزوپلی ساکارید تولیدی هر کدام از جدایهها در غلظت 5 % نمک تعیین و سویه برتر مورد شناسایی قرار گرفت. ماهیت اگزو پلی ساکارید سویه برتر با استفاده از تکنیک FTIR و مقدار اگزو پلی ساکارید تولیدی در غلظتهای بالاتر نمک نیز با روش آنترون تعیین گردید. در طیف مادون قرمز اگزوپلی ساکارید سویه مورد نظر پیکهای جذبی مربوط به ترکیبات کربوهیدراتی از جمله بی گلوکان، همچنین گروههای الکل، فنول، کربوکسیلیک اسید، کربونیل و آلکین مشاهده شد و تولید اگزوپلی ساکارید توسط جدایه فوق به طور معنیداری (05/0 > P) همراه با افزایش نمک، افزایش یافت. بر اساس نتایج سویهای با توان رشد در محیط 25 % نمک و تولید g/L 168/0 اگزوپلی ساکارید در مدت 24 ساعت به عنوان سویه برتر انتخاب شد و بر اساس توالیS rDNA 16 به عنوان سیتروباکتر فروندی سویه ATHM38 با شماره دسترسی KX553903 در پایگاه NCBI ثبت گردید.

اثر باکتریهای افزاینده رشد و قارچ میکوریزا بر رشد و عمکرد گندم در شرایط دیم
به منظور بررسی رشد و عمکرد گندم در شرایط دیم، آزمایشی مزرعهای به صورت فاکتوریل در قالب طرح بلوکهای کامل تصادفی با سه تکرار در مزرعه دانشکده کشاورزی، دانشگاه ایلام در سال زراعی 93-1392 اجرا شد. تیمارهای آزمایشی شامل عامل رقم گندم در دو سطح (کراسسبلان و ساجی) و عامل منابع کودی در هشت سطح شامل: 1- عدم مصرف کود شیمیایی فسفر، 2- 100 درصد کود شیمیایی فسفر، 3- باکتری سودوموناس پوتیدا (سویه 168)، 4- قارچ فانیلی فورمیس موسه، 5- باکتری سودوموناس پوتیدا+قارچ فانیلیفورمیس موسه، 6- باکتری سودوموناس پوتید+قارچ فانیلیفورمیس موسه+50 درصد کود شیمیایی فسفر، 7- باکتری سودوموناس پوتیدا+50 درصد کود شیمیایی فسفر و 8- قارچ فانیلی فورمیس موسه+50 درصد کود شیمیایی فسفر بودند. نتایج این پژوهش نشان داد که اثر رقم و منابع کودی بر وزن خشک سنبله، وزن خشک ساقه، میزان کلروفیل a و b، وزن خشک ریشه، مجموع طول ریشه، نیتروژن، فسفر، پتاسیم، طول دوره پرشدن دانه، سرعت پر شدن دانه، انتقال مجدد ماده خشک و میزان فتوسنتز جاری معنیدار بود. رقم ساجی دارای بیشترین سرعت پر شدن دانه، انتقال ماده خشک از ساقه، سنبله و فتوسنتز جاری بود. کود زیستی در شرایط دیم دارای اثر مثبت و معنیداری بر طول پرشدن دانه، سرعت پر شدن دانه، انتقال مجدد ماده خشک و میزان فتوسنتز جاری داشت، به طوری که در بین تیمارهای منابع کودی، قارچ فانیلی فورمیس موسه+50 درصد کود شیمیایی فسفر بیشترین طول دوره پرشدن دانه، سرعت پر شدن دانه، انتقال مجدد ماده خشک، میزان فتوسنتز جاری، میزان کلروفیل a و b، وزن خشک ریشه، مجموع طول ریشه و نیتروژن برگ را دار بود. نتایج این تحقیق نشان داد که در شرایط کشت گندم دیم در ایلام که گیاه با تنشهای آخر فصلی (خشکی و دما) مواجه میگردد، بنایراین گندم دیم رقم ساجی و قارچ فانیلی فورمیس موسه+50 درصد کود شیمیایی فسفر به دلیل بالا بودن میزان کلروفیل، عناصر غذایی، سرعت و طول دوره پر شدن دانه و فتوسنتز جاری که سبب افزایش عملکرد دانه میگردند را میتوان پیشنهاد نمود.

پایداری پادزیستی ازتوباکترها، انتروباکترها و سودوموناسها در خاکهای معدن، چراگاه و کشاورزی پیرامون سه معدن در استان همدان، ایران
فلزهای سنگین میتوانند سبب افزایش پایداری پادزیستی باکتریها شوند. هدف پژوهش بررسی پایداری پادزیستی باکتریهای گرم منفی در خاکهای دارای غلظت بالای فلز سنگین با کاربری معدن، چراگاه و کشاورزی بود. بنابراین 97 جدایه از 3 گروه باکتریهای گرم منفی غالب در این خاکها، برگزیده و پایداری آنها برابر 7 پادزیست و فلز سنگین بهروش پخشیدگی و ارزیابی MIC سنجیده شد. طبق دادهها 50 درصد ازتوباکترهای خاکهای کشاورزی و 6/28 درصد جدایهها در چراگاهها به آموکسیسیلین، آمپیسیلین، ونکومایسین و تتراسایکلین پایداری همزمان داشتند. 5/62 درصد انتروباکترهای خاکهای کشاورزی، 3/33 درصد در چراگاهها و 0/35 درصد در معدن به همین پادزیستها پایدار بودند. 5/87 درصد سودوموناسهای خاکهای کشاورزی و همه سودوموناسهای چراگاهها به این 4 پادزیست پایداری نشان دادند. پایداری به استرپتومایسین و داکسیسایکیلین در سه گروه باکتری، بالا بود اما پایداری به جنتامایسین کمترین بود. همه جدایهها به سرب پایدار و به کادمیوم ناپایدار بودند. %100 ازتوباکترهای خاکهای کشاورزی، بیش از 0/57 درصد ازتوباکترهای چراگاهها و 7/66 درصد ازتوباکترهای خاک معدنها به سرب، جیوه و نیکل پایداری همزمان داشتند. فراوانی انتروباکترهای پایدار به این فلزها در سه کاربری کشاورزی، چراگاه و خاک معدن 8/45، 0/80 و 0/65 درصد بود. همچنین 5/37 و 7/66 درصد سودوموناسهای خاکهای کشاورزی و معدن به این فلزها پایدار بودند. پایداری پادزیستی باکتریها میتواند بهدلیل غلظت بالای فلزهای خاک باشد. پایداری پادزیستی باکتریها در همه کاربریها بالا بود اما خاکهای کشاورزی زیستگاه شایستهتری برای افزایش باکتریها هستند و خطر انتقال باکتریهای پایدار و بهدنبال آن ژنهای پایداری از این راه به چرخه غذایی نیز بیشتر است.

بررسی تأثیر باکتری حل کننده فسفات، ورمی کمپوست و منابع فسفات بر رشد کاهو در یک خاک آهکی
در سالهای اخیر استفاده از منابع نامحلول فسفات به همراه کودهای زیستی در خاکهای آهکی به طور قابل توجهی افزایش یافته است. به منظور بررسی اثر باکتری حل کننده فسفات، ورمیکمپوست و منابع فسفات بر برخی ویژگیهای رویشی کاهو (رقم فردوس)، آزمایشی بهصورتفاکتوریلدرقالبطرحپایهکاملاًتصادفیدرسهتکراروباسهعامل انجام شد. عوامل آزمایش شامل باکتری در دو سطح (با و بدون مایه تلقیح)، منابع فسفات در چهار سطح (شاهد، خاک فسفات معدن آسفوردی یزد، تری کلسیم فسفات، و سوپر فسفات تریپل به میزان 25 میلیگرم فسفر در کیلوگرم خاک)، ورمیکمپوست در دو سطح (شاهد و یک درصد وزنی خاک) انجام شد. استفاده از کودهای زیستی موجب افزایش وزن تر و خشک اندام هوایی کاهو به طور معنادار شد هرچند که بر ارتفاع بوته کاهو اثر معناداری نداشتند. کاربرد خاک فسفات و تری کلسیم فسفات، به ترتیب موجب افزایش وزن خشک و وزن تر اندام هوایی کاهو گردید. کاربرد سوپر فسفات تریپل، وزن تر، وزن خشک و ارتفاع بوته کاهو را به طور معنادار افزایش داد. کاربرد همزمان باکتری، ورمیکمپوست و منابع فسفات (به جز سوپر فسفات) موجب افزایش وزن تر، وزن خشک، تعداد برگ و غلظت فسفر کاهو شد. بطورکلی نتایج نشان داد که کاربرد همزمان منابع نامحلول فسفات با کودهای زیستی میتواند جایگزین مصرف کودهای شیمیایی فسفاته گردد.
