Two experiments (pot and in vitro cultures) were conducted according to this hypothesis that heavy metals stress could increase glomalin production and metal complexes. Glomalin Bradford reactive extracted from hyphal compartment and the amounts of Pb-sequestrated by glomalin were investigated both in pot and in vitro cultures. Pot experiment was studied in washed sands using clover plant colonized by Rhizophagus irregularis fungus and treated with 0, 150, 300 and 450 µM of Pb. In vitro experiment was performed in two-compartments plates containing carrot roots colonized by the same fungus and treated with 0, 0.01, 0.1 and 1 mM of Pb as Pb(NO3)2. There was no significant correlation between mycorrhizal parameters including dry weight of fungus mycelium and percentage of hyphal frequency with content of glomalin extracted from hyphal compartment of pot and in vitro cultures, respectively. Glomalin contents extracted from hyphal compartment of pot and in vitro cultures were significantly improved by increasing Pb concentration compared to the control treatment. Pb-sequestrated by glomalin significantly increased as Pb concentrations and glomalin production increased. There was positive and significant correlation between Bradford reactive hyphal glomalin with Pb-sequestrated by glomalin. The results of this study revealed that the glomalin plays important role in Pb sequestration and thereby makes it valuable in phytostabilization of Pb in polluted soils.}, keywords = {arbuscular mycorrhizal fungi,Glomalin,Heavy metal and Phytostabilization}, title_fa = {تولید گلومالین توسط رایزوفاگوس ایرگولاریز در شرایط درون شیشه‏ای و کشت گلدانی شبدر سفید و نقش آن در غیرپویایی سرب}, abstract_fa = {گلومالین، به عنوان گلیکوپروتئین تولید شده توسط قارچ‏های میکوریزی در غیرپویایی فلزات سنگین در خاک اهمیت دارد و با توجه به این پیش فرض که تنش ناشی از فلز سنگین می‏تواند میزان تولید گلومالین و در پی آن فلز کمپلکس شده توسط آن را افزایش دهد، پژوهشی به ‏صورت گلدانی و درون شیشه‏ای طراحی گردید. مقدار گلومالین کل واکنش‏پذیر بردفورد استخراجی از بخش هیفی و مقدار سرب کمپلکس شده توسط آن در کشت گلدانی و درون شیشه‏ای مورد بررسی قرار گرفتند. آزمون گلدانی در بستر شنی به ‏همراه گیاه شبدر سفید میکوریزی‏شده با قارچ رایزوفاگوس ایرگولاریز و تیمار شده با چهار سطح 0، 150، 300 و 450 میکرومولار سرب انجام گرفت. آزمون درون شیشه‏ای در ظروف پتری دو بخشی حاوی ریشه‏های تراریخت هویج میکوریزی‏شده با همان قارچ و تیمار شده با چهار سطح 0، 01/0، 1/0 و 1 میلی‏مولار سرب از منبع نیترات سرب مورد بررسی قرار گرفت. همبستگی مشخصی بین شاخص‏های میکوریزی شامل وزن خشک هیف‏های خارج سلولی قارچ و درصد فراوانی هیف به ترتیب با میزان گلومالین تولیدی در بخش هیفی کشت گلدانی و درون شیشه‏ای مشاهده نگردید. همچنین با افزایش غلظت سرب، مقدار گلومالین استخراجی از بخش هیفی کشت گلدانی و درون شیشه‏ای به طور معنی‏داری نسبت به شاهد (فاقد تیمار سرب) افزایش یافت. مقدار سرب کمپلکس شده توسط گلومالین با افزایش سطح سرب و نیز افزایش تولید گلومالین به طور معنی‏داری افزایش یافت که بیانگر همبستگی مثبت و معنی‏دار بین گلومالین هیفی واکنش‏پذیر بردفورد با مقدار سرب کمپلکس‏شده توسط گلومالین می‏باشد. نتایج این مطالعه نقش گلومالین را در غیرپویایی سرب و اهمیت آن در استفاده از فناوری تثبیت گیاهی در خاک‏های آلوده به سرب نشان می‏د‏‏هد.}, keywords_fa = {تثبیت گیاهی,فلز سنگین,قارچ‏های میکوریز آربوسکولار و گلومالین}, url = {https://sbj.areeo.ac.ir/article_105960.html}, eprint = {https://sbj.areeo.ac.ir/article_105960_a7743bbda465921568c25cff90b3af6e.pdf} }
